I have a 2002 FXDWG3, that is mostly stock. Sometimes I lose spark at the rear cylinder, and to get spark I have to pull the wire off at the plug or the coil and then attach leaving about a eighth of an inch gap. I have checked both the coil an the wire an both checked good according to the service manual. Anybody got any Ideas?

make sure both ends are clean & with key off use a small wire brush to clean inside the coil , might be just a build up in there kinda like a battery gets & you have to clean them every once in a while

do you have chrome spark plug covers>?

They have rubber boots below the chrome to keep the covers
from grounding. If they slip, get a cut, etc then the plug can arc outside the cyl
    and it will missfire

Other cause can be a cut plug wire, you might have a slit in the wire and when you pull the wire back up it's closing the gap, inspect the wire from tip to tail...

Might consider replacing wires and i'd just use stock HD stuff, especially if you have the Delphi EFI as it has an ion sensor in the ecm and if you use the wrong plugs, wires etc it can toss off codes and cause issues.
